A challenge for the aquaculture community has long been the development of harmless techniques for monitoring fish in a tank. Acoustic telemetry has been used to monitor fish swimming behavior, and passive acoustics have been used to monitor fish feeding, but new techniques are needed to monitor non-invasively their numbers and growth rates. Recently, it has been demonstrated that the acoustical total scattering cross section of fish swimming in a tank can be measured from multiple reverberation time series. Rearing turbot (Scophthalmus maximus) reaches the commercial scale. With the hatchery output increasing, more attention needs to be focussed on broodstocks performance both in terms of quantity and viability of eggs. The techniques of shifting the environmental conditions allow a production of eggs all year round. However, egg quality tends to be highly variable. This paper examines over a two-year period, the improvement in egg quality and its consequences on the production of larvae by a strict management of spawners.

Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. Characterization of sperm motility in sea bass: the effect of heavy metals and physicochemical variables on sperm motility ArchiMer

Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Artificial insemination mastery in red drum, Sciaenops ocellatus, requires a first experimental step wOOng at getting basic biological data. Regarding males, a relation between optical density of the solution (OD) measured by spectrophotometry and sperm concentration (SC) has been established : OD O,161*SC + 0,119. Mean sperm concentration measured is 4,36.10 10 spz.mL-1. This study shows an influence of the solution of dilution on spermatozoa motility and its evolution during short term preservation. Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Cryopreservation of sperm in marine fish ArchiMer

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. Effect of stripping frequency on turbot spawn quality. ArchiMer

In production operations the application of daily examination of each female of broodstock should be a determinant progress in hatchery management in terms of fry output. The eggs expelled shortly after ovulation by means of a systematic fivefold a week stripping attempt of females, presented a high viability rate (72%) compared to that of eggs collected twice a week (46%) whereas the treatments did not have any effect on female fecundity. Increase of the stripping rhythm brought the hatching rates and broodstock fertilities respectively from 11% to 35% and from 20,000 to 65,000 larvae/kg of spawning female. The enhancement of egg quality and of the resulting larva production is discussed.

Effect of water temperature on individual reproductive activity of pollack (Pollachius pollachius) ArchiMer

The effects of water temperature on pollack (Pollachius pollachius) spawning features were individually assessed. During the spawning period, nine fish sets, each composed of one female and two males (mean body weight: 2.5+/-1.0 kg), were placed in small-volume tanks (2 m(3)). Fish sets were held at 8, 10, or 12 degreesC (n=3 for each temperature). Three females released eggs at 8 and 10 degreesC, while only two spawned at 12 degreesC. One hundred thirty-one individual spawns were collected from the eight spawning fish sets. The number of spawns collected per female was significantly lower at 12 degreesC (3.0+/-3.6), compared to 10 degreesC (17.3+/-10.1) and 8 degreesC (23.3+/-2.5). Egg production data from individual hand stripped female turbot were collected and analysed over several consecutive spawning seasons, on natural or shifted conditions. Egg quantity, egg viability and number of spawns were examined from one spawning season to the other, and inside the spawning season. These parameters seemed to increase from a year to the next one, except for the oldest fishes. Spawns were spread over a three-month period and occurred at the same time in the period in successive seasons for most females. Egg production seemed related to the feeding level and the earliness of spawning season appeared depended on the growth rate. Egg quality in domesticated and wild seabass (Dicentrarchus labrax): A proteomic analysis ArchiMer

In order to improve Our understanding of factors determining egg quality in fish, seabass egg batches obtained from 34 females held under standard conditions, were assessed for biological performances and a fraction from each batch was subjected to proteomic analysis. Comparison of proteomic patterns from eggs of different quality revealed differential protein spots among which one was identified as Nucleoside Diphosphate Kinase, a highly conserved protein involved in cell proliferation. This study shows that molecular markers may help understand factors determining egg quality in fish.
